Thymidylate kinase

Thymidylate kinase is a biology topic covered in the lgStudy science library. This page brings together a partial reference excerpt, illustrations, worked examples, real-world applications and a short study plan, so you can understand Thymidylate kinase rather than just read about it. In short: Thymidylate kinase (EC 2.7.4.9; dTMP kinase) catalyzes the phosphorylation of thymidine monophosphate (dTMP) to form thymidine diphosphate (dTDP) by transferring a phosphate group from the cofactor, adenosine triphosphate (ATP), which is converted to adenosine diphosphate (ADP): The enzyme characterised from Escherichia coli and mouse hepatoma requires magnesium ion Mg2+. Key takeaways

  • Thymidylate kinase belongs to biology; place it in that map before memorising details.
  • Learn the definition first, then one example that makes the definition concrete.
  • Connect Thymidylate kinase to a quantity you can measure, compute or draw — that is where exam questions come from.
  • Reproduce the core statement of Thymidylate kinase from memory before moving on to harder problems.

Reference excerpt

Thymidylate kinase (EC 2.7.4.9; dTMP kinase) catalyzes the phosphorylation of thymidine monophosphate (dTMP) to form thymidine diphosphate (dTDP) by transferring a phosphate group from the cofactor, adenosine triphosphate (ATP), which is converted to adenosine diphosphate (ADP):

The enzyme characterised from Escherichia coli and mouse hepatoma requires magnesium ion Mg2+. Thymidylate kinase is a ubiquitous enzyme of about 25 Kd and is important in the dTTP synthesis pathway for DNA synthesis. The function of dTMP kinase in eukaryotes comes from the study of a cell cycle mutant, cdc8, in Saccharomyces cerevisiae. Structural and functional analyses suggest that the cDNA codes for authentic human dTMP kinase. The mRNA levels and enzyme activities corresponded to cell cycle progression and cell growth stages. Thymidylate kinase's subfamily is predicted thymidylate kinase, TKRP1. InterPro: IPR014505 Human protein DTYMK contains this domain.

Structural studies As of late 2007, 40 structures have been solved for this class of enzymes, with PDB accession codes PDB: 1E2D, PDB: 1E2E, PDB: 1E2F, PDB: 1E2G, PDB: 1E2Q, PDB: 1E98, PDB: 1E99, PDB: 1E9A, PDB: 1E9B, PDB: 1E9C, PDB: 1E9D, PDB: 1E9E, PDB: 1E9F, PDB: 1G3U, PDB: 1GSI, PDB: 1GTV, PDB: 1MRN, PDB: 1MRS, PDB: 1N5I, PDB: 1N5J, PDB: 1N5K, PDB: 1N5L, PDB: 1NMX, PDB: 1NMY, PDB: 1NMZ, PDB: 1NN0, PDB: 1NN1, PDB: 1NN3, PDB: 1NN5, PDB: 1TMK, PDB: 1W2G, PDB: 1W2H, PDB: 2CCG, PDB: 2CCJ, PDB: 2CCK, PDB: 2PBR, PDB: 2TMK, PDB: 3TMK, PDB: 4TMK, and PDB: 5TMP.

Worked examples

Example 1 — a first encounter with Thymidylate kinase

Start with the simplest possible case. Write down what Thymidylate kinase claims or describes in one sentence, then invent the smallest concrete situation in which that sentence is true. In biology, the smallest case is usually a single object, a single equation or a single measurement. Check that every symbol or term in your sentence has a meaning in that case.

Example 2 — changing one variable

Take the situation from Example 1 and change exactly one quantity: double it, halve it, or set it to zero. Predict what should happen to Thymidylate kinase before you calculate. Comparing your prediction with the result is the fastest way to find out whether you understand the idea or only the words.

Example 3 — an exam-style question

Typical questions about Thymidylate kinase ask you to (a) state it precisely, (b) apply it to given data, and (c) explain a limitation. Practise writing all three answers in under five minutes; the third part is what separates a full-mark answer from an average one.

Applications of Thymidylate kinase

In research
Thymidylate kinase appears in biology research whenever the underlying quantities have to be modelled precisely. Papers usually cite it as a starting assumption and then explore where it breaks down.
In technology and industry
Engineering practice reuses Thymidylate kinase in design rules, simulations and safety margins. Knowing the idea lets you read a specification sheet and understand why the numbers look the way they do.
In the classroom
Thymidylate kinase is common in secondary-school and first-year university syllabi. It links to neighbouring topics EC 2.7.4, Enzymes of known structure, Protein families, so understanding it makes those chapters shorter.
In everyday life
Look for Thymidylate kinase outside the textbook — in sport, cooking, traffic, electronics or the sky above you. An example you found yourself is remembered far longer than one you were given.

Frequently asked questions

What is Thymidylate kinase in simple terms?

Thymidylate kinase (EC 2.7.4.9; dTMP kinase) catalyzes the phosphorylation of thymidine monophosphate (dTMP) to form thymidine diphosphate (dTDP) by transferring a phosphate group from the cofactor, adenosine triphosphate (ATP), which is converted to adenosine diphosphate (ADP): The enzyme characte…

Why does Thymidylate kinase matter?

Because it connects several biology ideas at once: it gives you a definition you can apply, a quantity you can calculate, and a way to check whether a result is plausible.

How should I study Thymidylate kinase?

Read the excerpt, restate it from memory, then work through the examples and applications listed on this page. The five-step study plan above takes about twenty minutes.

What does this page cover?

It gives you a compact reference excerpt plus original lgStudy explanations, examples, applications and study material on Thymidylate kinase.
